The purification of the enzyme recently received increasing attention because the enzyme was used to determined the carbonhydrate structures of oligo-mannoproteins, which has specific biological activities. An α-mannosidase was purified over 100-fold from Wata callus by successive chromatography with overall yield of 8%. The purified enzyme had a molecular mass of 250 kDa. This enzyme had the same optimum pH at 4.5 and optimum temperature at 50℃ as one from jack bean. This enzyme appeared to be metal enzyme containing Zn^<2+>. The enzyme hydrolyzed p-nitrophenyl- α -mannoside, methyl- α -n-mannopyranoside, benzyl- α -Dmannopyranoside, α(1->2)-mannobiose, α(1->3)-mannobiose, and α(1->6)-mannobiose, with Km of 0.527mM, 0.182mM, 0.190mM, 1.06mM, 0.696mM, 5.10mM, respectively The hydrolysis of various α-linked mannobiose indicated that the enzyme hydrolysizes the α-mannobiose in the order of α(1->6) > α(1->3) > α(1->2), unlike the conventional α-mannosidae.","subitem_description_type":"Abstract"},{"subitem_description":"近年,生体細胞の細胞表面あるいは酵素表面に結合した糖鎖が,細胞の生理活性あるいは酵素活性に多大な影響を示すことが報告されている。例えば,maltoseを加水分解するヒトの腸間酵素は重量に対して30-40% の糖を含んでいるが,papainの加水分解に対して抵抗性を示す。一方,酵母の細胞壁と結合したmannoseを多量に含む糖鎖は,酵母の性的凝集反応を引き起こすことが知られている。特に,Saccharomyces cerevisiaeにおける糖鎖の構造は古くから検討されており,糖鎖の構造は,mannoseがα(1→6)結合で結合したoligo-mannose骨格に,側鎖として2,3個のmannoseがα(1→2),または,α(1→3)結合したmannobioseとmannotrioseが結合されていると報告されている。近年,この糖タンパクの生物学的役割を明らかにするため,oligo-mannose型糖鎖の構造を解明することが重要となってきた。そこで,この糖鎖の結合状態を決定するために,特定の結合部位のみを加水分解することが可能なα-mannosidaseが必要とされている。現在まで,jack beanから精製したα-mannosidaseが安価で入手しやすいことから,oligo-mannoseの構造決定に使われてきた。しかし,この酵素は加水分解速度が遅く,さらに,基質特異性としてα(1→2),α(1→3),α(1→6) 結合の順に切断するため,mannose含量を決定するために利用できるが,明確な構造決定に利用できなかった。また,近年,発見された微生物由来のα-mannosidase は,主にα(1→2),α(1→3)結合を特異的に加水分解すると報告されている。従って,oligo-mannose型糖質の構造決定を行うために新しい切断特性を有するα-mannosidaseの検索とその性質を決定することが急務となってきている。そこで我々の研究室では新しい起源からα-mannosidaseを生産回収することを目的として担子菌植物などを用いて検索した結果,綿カルスの細胞内にα-mannosidaseを高濃度に蓄積することを発見した。本研究では,この綿カルスからα-mannosidaseを高純度に精製することを検討した。さらに,精製した酵素の至適pH,反応速度パラメーター,基質特異性等の特性を,従来報告されているα-mannosidaseと比較検討した。","subitem_description_type":"Abstract"}]},"item_3_description_40":{"attribute_name":"資源タイプ(DSpace)","attribute_value_mlt":[{"subitem_description":"Article","subitem_description_type":"Other"}]},"item_3_description_6":{"attribute_name":"引用","attribute_value_mlt":[{"subitem_description":"富山大学工学部紀要,48,
